Full description or abstract By using ECM proteomics we have defined the matrisome of dormant cancer cells vs proliferative cancer cells of head and neck squamous cell carcinoma (HNSCC). To do so, we used established cellular dormancy models and their proliferative counterparts: proliferative (T-HEp3) and dormant (D-HEp3) HNSCC. To investigate the matrisome composition of these dormant and proliferative tumors, we performed ECM-enriched mass spectrometry using decellularized T-HEp3 tumors and D-HEp3 dormant nodules grown in mice. Analysis of indolent D-HEp3 dormant nodules and aggressive T-HEp3 tumors grown in mice show a significant dysregulation of the matrisome signature. This gene set lists the matrisome proteins detected exclusively in highly proliferative HNSCC human-to-mouse xenograft compared to dormant HNSCC human-to-mouse xenograft. We further excluded proteins that were detected in only one of the three proliferative samples.
