Full description or abstract Drug-tolerance has emerged as one of the major non-genetic adaptive processes driving resistance to targeted therapies such as tyrosine kinase inhibitors (TKI) in non-small cell lung cancer (NSCLC). To identify potential hallmarks of drug-tolerant cells (DTC), we performed RNAseq and scRNAseq experiments using different models of EGFR-mutant DTC that emerged in response to EGFR-TKI (erlotinib or osimertinib) after 7-to-21 days of treatment. We combined our transcriptomic data with publicly available resources to generate a signature of drug-tolerance consisting in the most commonly overexpressed (p<0.01, Log2FC>0.5) or downregulated (p<0.01, Log2FC<0.5) genes in at least 6 out of 7 models.
