Brief description The NRG (Necroptosis-Related Gene) signature is an 18-gene transcriptional model designed for the prognostic stratification and immunological profiling of colorectal cancer (CRC).
Full description or abstract The NRG signature effectively classifies patients into high-risk and low-risk categories based on a calculated risk score. The low-risk group is significantly associated with an immune hot phenotype, characterized by high infiltration of cytotoxic T cells, NK cells, and the upregulation of 14 HLA-related antigens, as well as higher immune checkpoint activity. The high-risk group reflects an immune cold microenvironment with lower overall immune and TIDE scores. The signature's prognostic accuracy was validated across three independent cohorts (TCGA-COAD, GSE38832, and GSE161158), where a nomogram integrating the NRG score with AJCC staging achieved an AUC of 0.811 for 5-year overall survival.
