Full description or abstract This gene set represents a 24-gene telomere-related gene (TRG) signature developed to predict the prognosis and immune landscape of head and neck squamous cell carcinoma (HNSCC). Using machine learning on data from TCGA and GEO, researchers established a risk-scoring model that classifies patients into high- and low-risk categories. The signature demonstrated strong predictive accuracy, with 3-year and 5-year survival ROC AUC values of 0.734 and 0.711, respectively. Patients in the low-risk group exhibited significantly better survival outcomes, characterized by higher immune scores, increased immune cell infiltration, and a superior response to immunotherapy. Furthermore, through Mendelian randomization, the study identified a potential causal link between MAD1L1 expression and HNSCC development.
