Full description or abstract This gene set represents a consensus senescent cancer-associated fibroblast (senCAF) signature derived from two independent studies of stromal senescence in breast cancer (Ye et al., Cancer Discovery, 2024; Belle et al., Cancer Discovery, 2024) and bresat bone metastasis (Faget et al., Cancer Discovery 2023) . Genes were extracted from CAF-adapted senescence modules in each study and merged with deduplication. The signature captures core senescence markers (CDKN1A, CDKN2A), SASP cytokines (IL6, IL8, CCL2, CXCL1/2/3), stress-response effectors (ATF3, HSPB1, DUSP1, ZFP36 family), ECM/fibrotic remodeling genes (COL1A1/1A2, COL12A1, FN1, THBS1/2), and senescence-associated osteogenic and immunomodulatory factors (SPP1, TGFB3, CD276, TNFSF11). This integrated signature reflects shared, reproducible features of stromal senescence across datasets and can be used for GSEA, ssGSEA, and stromal senescence scoring in bulk or single-cell transcriptomic data.
